Beyond probiotics: the next gut supplement that researchers are watching closely

Postbiotics, the beneficial compounds derived from gut bacteria rather than the live bacteria themselves, are the next wave of gut health products moving from labs onto supplement shelves. A 2025 review in Frontiers in Microbiomes describes them as bacterial byproducts and cell fragments that appear to influence gut and immune health without needing to be alive.

The category covers short-chain fatty acids such as butyrate, small proteins called bioactive peptides, complex sugars produced during fermentation, and pieces of bacterial cell walls. In 2021, an international scientific panel writing in Nature Reviews Gastroenterology and Hepatology formally defined a postbiotic as "a preparation of inanimate microorganisms and/or their components that confers a health benefit on the host."

That difference from probiotics matters in practice. Postbiotics do not need to survive shelf storage or stomach acid, are more stable in products, and, because they contain no living organisms, they sidestep the question of live bacteria interacting with the gut in people with weakened immune systems.

What the research shows

Two 2025 reviews point in a similar direction. The Frontiers in Microbiomes paper concluded that postbiotics support gut barrier function, immune modulation, and antioxidant activity, with early evidence of a role in managing obesity, diabetes, and cardiovascular disease.

A separate review published in May 2025 in the Journal of Clinical Medicine found postbiotics strengthen the intestinal wall through several mechanisms. These include boosting tight junction proteins, known by names such as ZO-1, occludin, and claudin-1, which seal the space between gut cells.

On the immune side, the same review reported that postbiotics can shift the body toward anti-inflammatory signaling, raising levels of the calming cytokine IL-10 while lowering markers such as TNF-alpha and IL-6.

Blood sugar effects are less clear-cut. Short-chain fatty acids produced by gut bacteria are known to affect glucose and lipid metabolism, but robust human trials testing postbiotic products specifically for blood sugar control remain limited.

What's not settled yet

The evidence base is still early. Most findings come from cell studies, animal experiments, and small human trials rather than large clinical studies. The Frontiers authors call for more comprehensive safety evaluations, particularly in allergic and immunocompromised populations.

Regulation is also playing catch-up. There is no approved postbiotic drug and no dietary intake target, and the amounts of postbiotic compounds in different fermented foods and supplement products vary widely.

For readers already taking a probiotic, the practical takeaway is measured. Postbiotics look like a promising second wave rather than a proven treatment, and they are not yet backed by the kind of large, long-term trials that would justify a change in medical care. Anyone considering a new supplement, especially with a chronic condition or a weakened immune system, should raise it with their doctor first.
